1 Wnt4 重组蛋白对鸡等级卵泡颗粒细胞相关基因表达的影响摘 要鸡的卵泡选择与产蛋量高度相关,研究卵泡选择的分子机制可为提高其产蛋性能提供一定的理论依据。Wnt 信号通路能够调控很多卵巢生长发育过程中的生理过程,如卵泡发育、排卵和黄体化。Wnt4 是 Wnt 家族的成员,对雌性动物的生殖系统起到一定的调控作用。为了研究 Wnt4 对鸡卵泡选择的调控作用,本研究检测了 Wnt4 重组蛋白对鸡等级卵泡颗粒细胞中相关基因表达的影响作用,结果发现 Wnt4 重组蛋白能够显著上调 FSHR 和 CTNNB1的表达,并对 StAR 和 CYP11A1 的表达有一定的促进作用。本研究的结果可为深入研究 Wnt4的具体功能提供一定的研究基础,也可为进一步阐明鸡卵泡选择的分子机制提供参考。关键词:鸡;卵泡选择;Wnt 信号通路;Wnt4;qRT-PCREffect of Wnt4 Recombinant Protein on Expression of Genes Related to Granulosa Cells of Chicken hierarchal Follicle 2ABSTRACTOvarian follicle selection is an important process impacting the fecundity of hens. Studying the molecular mechanism of follicle selection can provide a theoretical basis for improving egg production performance. Wnt signaling pathway can regulate many physiological processes during ovarian growth and development, such as follicle development, ovulation and luteinization. Wnt4 is a member of the Wnt family , it plays a regulatory role in the reproductive system of female animals. In order to study the effect of Wnt4 on follicle selection, this study examined the effect of Wnt4 recombinant protein on the expression of related genes in chicken hierarchal follicle granulosa cells. The results showed that Wnt4 recombinant protein could significantly increased the expression of FSHR and CTNNB1, it also could promote the expression of StAR and CYP11A1.
