脱细胞支架的胎儿细胞注射 摘要:建立体外心肌细胞分离培养、纯化和鉴定方法。[方法]若干只小鼠分 10 次采用分离消化法培养心肌细胞,分别用 A、B、C、D 4 组不同培养液培养,以 Ficoll 梯度液纯化,进行 Desmin 免疫组化鉴定。[结果]心肌细胞培养后经台盼蓝检查成活率达 95%以上,在接种后 4~5 d 增殖达高峰,A、B、C 3 组培养液对心肌细胞的培养效果无差异。Ficoll 分离纯化的心肌细胞纯度较高,有 97%的心肌细胞 Desmin 胞浆呈阳性反应。[结果]采用 Ficoll 纯化以国产血清培养,可获得高产量高纯度的心肌细胞。关键词:脱细胞;支架;胎儿细胞From fetal cells were injectedAbstract: To establish a practical met hods of t he culture,purification and identification of mouse myoblast s in vit ro.Met hods:Muscle samples of 40 mice were minced and digested wit h t rypsin,cul2t ured wit h four kinds of media namely A,B,C,and D,finally purified wit h Ficoll gradient liquid.Myogenesis were dynamically observed,myoblast were tested by Desmin immunohistochemist ry stains.Result:The my 2 oblast cell viability is up to 95%by Trypan blue,t he cell proliferation arrived climax during 4 ~ 5 day after planted.There is no difference Ibetween A,B,and C cult ure liquid.The cell purity is up to 97%by Desmin im 2 munohistochemical stains test.Conclusion:Myoblast s could be purified by Ficoll and cult ured wit h DM EM cul2 t ure liquid contained serum made in our count ry,in high quantity.Key words:Take off, Stents, Fetal cellsIIIII1 引言目前全世界有2200万人感染了心脏衰竭。在同种异体移植中,已有肾、肝、心、肺、小肠、骨关节等移植成功的报道。但同种异体组织、器官仍有供者严重不足的困难。因此,寻找异种移植源成为现代移植外科研究的前沿。异种移植与同种异体移植一样,克服排斥反应将是今后研究的重点,但实质器官如心、肝、肾的异种移植一旦失败,则意味着患者丧失生命。心脏移植仍然是最终确定的治疗心脏衰竭阶段,但捐赠器官的供应是有限的。一旦心脏移植后,往往需要以交换高血压,糖尿病和肾衰竭...
